丸浜伸一朗先生という平成17年卒の大学院１回生ですが、現在当科講師の澤本伸克先生先生の直接指導のもとパーキンソン病の Neuromelanin MRIイメージングと病理対応というテーマで研究を開始しております。
MRI撮像は順調に立ち上がり、ある程度残存黒質ニューロン数や左右差を反映しているデータが得られつつ有りますが、その厚みを病理対応させ るのが以下の理由で困難となっております。

１）肉眼の中脳黒質割面の色調はドパミンニューロンの総数の評価には耐えるが、左右差は全く反映しない（逆の場合もある）。
２）通常の薄切切片では、頭尾方向の神経細胞の位置のズレのため、数枚おきに検鏡するにしても正確な評価が困難。
３）厚切り浮遊切片の染色で蛍光抗体なり、DAB抗体なり染めるにしても、やはり検鏡の際の再構築でバイアスが入る。

我々のラボミーティングで上記が議論となったのですが、丁度今年遠山先生が会長をされた細胞組織化学学会の講習会のテキストでJean- Pierreが脳透明化のハンズオンレクチャーをしていることを知りました。それを見まして、厚切り切片の脳透明化とTH染色を合わせた３次 元イメージが出来ればMRIとの対応がうまくいくのではと愚考いたしご相談させて頂いた次第です。丸浜先生と澤本先生にお話した所、大変興味を示されました。

ヒト脳ではまだまだハードルが高いかとは思いますが、試みる価値はあるプロジェクトと考えており、是非ご指導を頂けますと幸いです。本人は組 織化学の経験がありませんので、まずは動物脳から技術が身につくまで滋賀医大で技術習得をさせたいと思っております。

一度プロジェクトについて、丸浜先生に滋賀医大でプレゼントご説明をさせて頂き、可能かどうかご検討を頂けますと幸いです。
